Is it better/recommended to shut down your laptop whenever you throw it into the backpack and go on the move?
Or is it fine to leave it on (sleeping)?

Your supposed to shut it off, as hard drive spindown can cause problems, but I lugged my laptop around for 2 years with it just asleep and it never had any problems.

I would usually just put mines to sleep for the 30 - 45 minute commute. No problems.

you shouldn't use a backpack. laptops get all claustrophobic like a motherfucker

moving around a hard drive that is active can damage it...I would say no, dont do it.
